Synergistic anti-tumour effects of 13-cis retinoic acid and lovastatin in pancreatic neuroendocrine tumour (BON1) cells through enhanced EGFR inhibition

Nolting Svenja , Prada Elke Tatjana Aristizabal , Lauseker Michael , Maurer Julian , Spottl Gerald , Goke Burkhard , Pacak Karel , Grossman Ashley

Introduction: In our previous studies we found that the combination of 13-cis retinoic acid (13cRA) and lovastatin significantly reduced tumour growth in a mouse phaeochromocytoma allograft model, with the lowest microvessel density in the combination-treated tumours. We have now investigated the effect of 13cRA plus lovastatin on neuroendocrine (BON1, H727) and non-endocrine tumour (HepG2, Huh7) cell viability and signalling pathways (EGFR, AKT, ERK, p70S6K) to eluci...

Adrenocortical carcinoma (ACC) is rare malignancy associated with poor prognosis. The aim of this study was to present clinical characteristics and survival of patients with ACC and to analyse the effect of prognostic factors on survival. We retrospectively analysed 69 patients (46 female and 23 males) with pathologically confirmed ACC who were treated in our hospital between January 2005 and December 2015. The median age at diagnosis was 51,0 years (rang...

Introduction: During pregnancy (state with physiological hyperprolactinemia-HP) mothers require larger calcium amount for fetal growth. It has been shown that PRL stimulate intestinal calcium absorption in physiological HP, which can protect mother’s skeletal system from further mineral density loss. On the other hand, medicamentous-related hyperprolactinemia is more frequently associated with reduced BMD and increased fracture risk. We conducted the experimental study to...

Correlation of chromogranin a and standard metabolic parameters in patients with gastroenteropancreatic neuroendocrine tumors (GEP-NETs)

Bozic-Antic Ivana , Ognjanovic Sanja , Popovic Bojana , Isailovic Tatjana , Bogavac Tamara , Ilic Dusan , Elezovic-Kovacevic Valentina , Macut Djuro

Introduction: Chromogranin A (CgA) is a major representative of the protein family named granins stored predominately in secretory granules of the diffuse neuroendocrine system. Circulating CgA serves as a biomarker of neuroendocrine tumors (NETs), but its increased plasma concentration is present in various cardiovascular, gastrointestinal, renal and inflammatory diseases. Intra-granular and/or extracellular proteolysis of CgA generates a variety of biologically active fragme...

Changes in GLUT1 expression under metformin treatment in rat models of severe and mild stretozotocin diabetes mellitus

Sokolovska Jelizaveta , Isajevs Sergejs , Sugoka Olga , Sharipova Jelena , Lauberte Lasma , Svirina Darja , Rostoka Evita , Sjakste Tatjana , Kalvinsh Ivars , Sjakste Nikolajs

Introduction: Metformin improves hyperglycaemia via mechanisms which include activation of AMP-activated protein kinase (AMPK). Recent findings indicate that some metabolic actions of metformin occur also by AMPK-independent mechanisms. Objective: To study the action of metformin on expression of GLUT1 glucose transporter in rat models of severe and mild stretozotocin diabetes mellitus.Materials and methods: Severe diabetes mellitus...

Ghrelin is a somatotrophic, orexigenic and adipogenic hormone that has an important homeostatic role by linking regulatory systems for growth and energy balance. Ghrelin- secreting neuroendocrine tumors are rare, but we had the opportunity to study a 71 years old female patient with a gastric ghrelinoma. The patient was submitted to gastroenterological examination after having episodes of diarrhea, and small subepithelial polyp (5 mm) was revealed on gastroscopy. Pathohystolog...

Expansion of grey and white matter volumes and increased white matter gliosis in acromegaly: a clinicoradiological study of 43 patients

Sievers Caroline , Samann Philipp , Dose Tatjana , Czisch Michael , Dimopoulou Christina , Schneider Harald , Rommler Josefine , Schopohl Jochen , Stalla Gunter Karl

Background: In acromegaly, few in vivo data exist on changes of the brain parenchyma as a pathology caused by systemically elevated levels of GH and IGF-1. This is in contrast to neuropsychiatric symptoms such as depression, cognitive impairment, personality changes and pain syndromes that accompany the condition. We aimed at measuring brain tissue volumes and white matter pathology in acromegaly. Methods: Forty-three patients (age 54±14 years...

Screening for mutations in exon 10, 11, 13 and 14 of the RET protooncogene associated with inherited medullary thiroid carcinoma (MTC) in Serbian population

Nikolic Jelena , Vignjevic Jovana , Mirkovic Katarina , Beleslin-Cokic Bojana , Antic Jadranka , Isailovic Tatjana , Petakov Milan , Damjanovic Svetozar

Background: Ret protooncogene germ-line mutations are associated with the inherited multiple endocrine neoplasia type 2 syndromes (MEN2a and MEN2b) and also with familial medullary thyroid carcinoma (FMTC). In this study, we report a large scale of mutations in exon 10, 11, 13 and 14 RET protoocogene in patients from Serbia. Our study included patients with MTC.Methods: Our study included 180 patients. Patients were tested for RET protooncogene mutations...

Composite medullary and papillary tumor with mixed lymph node metastases

Isailovic Tatjana , Petakov Milan , Macut Djuro , Popovic Bojana , Bozic Ivana , Ognjanovic Sanja , Nikolic Jelena , Vignjevic Jovana , Mirkovic Katarina , Damjanovic Svetozar

A 34 year-old female patient was admitted to the hospital because of a large nodule in the left thyroid lobe and elevated calcitonine level. A large encapsulated tumor was found and total thyroidectomy with left neck dissection was performed. Pathohistology revealed medullary and papillary carcinoma separate from each other in tumor tissue but mixed in regional lymph nodes. Papillary component was dominant in thyroid tissue but not in lymph nodes. Both calcitonine and thyroglo...
